Release Notes 2.7.0

This new release of Fusion introduces modularity to the software and the implementation of several new features that increase user productivity.   

New modules (z intensity compensation, Focus Map and irregular Montage) can be purchased at any time to expand Fusion capabilities. Once a module is purchased, subsequent upgrades to this feature will be automatically included for clients who are under a maintenance contract. 

 

New Features- Included on Core Fusion 2.7 

Spiral Montage (Preview)    

  • Designed to allow quick sample exploration before protocol acquisition. 

  • Support preview and protocol montage that operate from the centre out in a spiral pattern. 

  • When clicked more than once, the Quick Tile Montage will add new fields to the existing preview image (creating a new image) 

  • Running Quick Tile at the same point multiple times will expand the area previewed at that position. 

  • Quick tile montage will only acquire tiles from the preview that had not yet been acquired (minimising acquisition time, storage requirements and sample bleaching). 

  • Moving the stage to a new area allows adding and clicking on the spiral montage icon, delivering the build-up of the pre-existing montage by adding the tiles of the new selected area.  

  • The preview image can be redisplayed after a protocol or snap is acquired. 

  • The preview image can be cleared, allowing the rebuilding of a new one. 

  • Preview images can be viewed on the file manager under “overlays”. 

  • Preview images can be used for sample navigation and selection of image acquisition area. 

 

Sample Exploration 

  • New sample navigation “eye” Icon allows the user to visualise where in the preview montage the stage is.   

  • If "Click To Navigate" is in use, the new “eye” Icon will outline the current stage position relative to the the image 

 

Stitching 

  • Added option to not align in tiles in Z when stitching. This option avoids blank tiles and additional z planes being inserted when stitching tilted samples.  

  • This option should be selected when stitching images acquired with the focus map feature.  

 

Montage Improvements  

  • When a Multi-Field is stopped, the fields that were not written to will not now appear in the dataset. 

  • When a Montage acquisition is stopped, the fields that were not written to will not now appear in the dataset 

 

Others  

  • Default image compression type set to 'LZ4Shuffle'.  

  • Multi-Field view mode, now supporting image panning and zoom to point 

  • A Position Interval can be set for multi position protocols to specify a minimum time between the start of a stage move and when imaging occurs at the new location.

New Features- Not Included on Core License

Focus Map (License Required) 

  • Focus Map feature allows the correction for Tilt or Focus Map (sample wobbling) 

  • Correction for Tilt in different sample holders.  

  • Correction for Tilt in Multiwell plates  

  • Focus map support – global for single position correction  

  • Focus map support – local for multiple positions correction  

  • Local Focus map – can be applied to each well in Multiwell corrected plates.  

  • Added a button allowing all local focus maps to be deleted at once. 

  • Tilt is corrected by introducing 3 points in the montage. 

  • Focus map is corrected by introducing 5 or more points in the montage. 

  • Complex focus map surfaces can be defined using multiple support points. 

  • Presents a notification when focus map configuration has too few points. 

  • Add Focus Map Positions via one-click on the display. 

  • A select Focus Map position can be updated via click on display 

  • Possible to add to the Focus map support points in the configuration table through stage navigation.  

  • Quick Review of Focus Map points. 

  • Includes a toggle button added to the Focus Map Specification Panel to show/hide positions on display. 

 

Z Intensity Correction (license required for BC43 Widefield) 

  • Allow the Laser power to be increased through a z-stack. 

  • During a Z scan, if Z intensity correction is enabled, laser power increases as we image deeper into the sample.   

  • Z Intensity compensation can be corrected by using either an exponential or a linear calibration function 

  • The user can select the calibration function that is more appropriate to the experiment.  

  • Z Intensity correction range can be set to a different start and end position than the Z scan range. 

  • Z Intensity Correction can be used with multiple channels 

  • Different channels can use a different intensity for Z intensity correction. 

  • Image Auto Mapping is automatically switched off when calibrating for laser intensity correction. 

 

Irregular Montage (License Required) 

  • Irregular montage (Perimeter) can be specified via a new drawing mode. 

  • The outline of the Perimeter Montage can be easily updated via an overlay on the main display.  

  • The outline (of the tiles to be acquired) of the specified Perimeter Montage can be viewed.   

  • New User interface allowing easy toggle between specifying the montage by Perimeter or tiles. 

  • Renamed the previous Fields montage to tiles  

  • Renamed the previous Edge montage to Perimeter.  

  • Perimeter Montage includes all fields that fall within the shape as defined by the specified positions of the 'irregular' montage shapes.  

  • If no irregular montage license is valid, this mode includes all fields that fall between the min and max positions of the specified positions. 

  

Bugs Fixes 

  • Fix crash when select an AOI by drawing on image that is larger than the camera sensor size. 

  • Fix montage tiles appearing in groups instead of individually during an acquisition when using Multi-Field view mode 

  • Fix single auto-map click button not updating the mapping 

  • Fix Image Intensity mean was showing incorrectly in Region Intensity Histogram Statistics. 

  • Fix auto-mapping only occurs when option is selected. 

  • Click To Navigate now operating correctly when viewing a data set with multiple fields, when displaying all fields
