Release 2.6.0

New Features:

Mosaic Support 

  • Device detection (Mosaic) 

    • Automatic detection of Mosaic device from Device Hub & Display information device in Device Hub

    • Detection and control of mosaic diode laser

    • Control of mosaic illumination area includes full area and bit map

  • Calibration

    • Fully automated 'one click' calibration of Mosaic device

    • Automatically detect the usable illumination area of the Mosaic and visualise to user using photostimulation bounds and toggle on/off

      • Regions that fall partially or wholly outside the Mosaic illumination area will be clipped to only illuminate the portion within the field

    • Calibration applies at the Mosaic device level and may be used for both:  1) camera used for the calibration and 2) the other camera in the system.

Regions and region management

  • Photostimulation region management

o    Regions can be tagged as either photostimulation or analysis.

o    New Overlay manager, which includes possibility to 1)  save load and export regions, 2) rename regions

o    Photostimulated overlays appear as recorded regions on the acquired image. These can be cloned as Analysis and/or Photostimulation overlay types for reuse

o    Recorded Regions are shown for each associated photostimulation event so it is clear where and when photostimulation took place during the acquisition of image data ,and can be toggled on/off for visualisation.

o    Support for saving the acquired time point / frame position to .roi files for use in Fiji

 

Photo Stimulation protocols

  • Trigger

    • Mosaic illumination time can be configured in channel settings

    • Mosaic device can be illuminated as part of a protocol trigger

    • Trigger can be configured to 1) fire at equally spaced time points after the initial trigger time point of the protocol, 2) add delay in the protocol.

    • BOB Output 6 can be used as a trigger in the protocol. (The spare output on the Bob device can be configured to pulse at the start of a specified timepoint during a protocol)

  • Manual Triggering

    • All protocol triggers can be manually fired with one click if desired

    • Each trigger that is configured in the active protocol can be triggered manually on demand

    • Allow Automatically move (skip) a running protocol to the next time point if the user manually fires one of its triggers

  • Regions within protocols

    • Phostimulation protocols with mosaic are possible and require the use of phostimulation regions and triggers.

  • Review photostimulation protocol

    •  The timepoints at which photo stimulation occurred is displayed on the image navigation bar

    •   Region manager panel added this lists all current regions with name and type

 

  Analysis

  • Through series analysis supports overlays.

  • Through series analysis has a progress bar on the start/stop button.

  •   Through series analysis will automatically switch to Time Series or Z Scan where applicable.

 

  Drift Stabilisation 

  •   Ability to automatically remove drift stabilisation dichroic mirror during a protocol for supported devices

  • Move Dichroic Mirror Out when Drift Disabled for Leica DMi8

  • Move Dichroic Mirror Out when Drift Disabled for Nikon Ti2

  • New protocol setting to control whether dichroic moves out when drift is disabled 

  •  Support removing drift stabilisation dichroic mirror in time series for Leica DMi8

  •  Support removing drift stabilisation dichroic mirror in time series for Nikon Ti2 

  • Updated drift stabilisation tooltips to be more informative

 

Other Improvements

  • Adds the ability to play sounds at key events during Fusions runtime

  • Added some 3rd party libraries to the Acknowledgements dialog

 

Bug Fixes

  • Fixed a crash related to trying to validate devices that have not finished connecting

  • Fixed crash when running a quick montage if the Montage bounds were not set.

  • Fixed Snap Live to ignore different channel settings.

  • Fixed Updating Z range using slider does to update start/end values properly

  • Fixed issue where clicking on a deconvolved image would cause the live images to overwrite the previous images.

  • Fixed deconvolution parameters reset on acquisition.

  • Fixed Image zoom on the mouse location when using the scroll wheel.

  • Fixed issue where Position and Intensity were not updated on Zoom in/out event when mouse not moved.

  • Fixed issue when position and intensity was not updated on zoom in/out on image display.

  • Scroll bars are now enabled when Fusion is Acquiring or Live 

  • Fixed "save image" dialog appearing when going Live, if user double clicked during a previous acquisition

 

Limitations to Photostimulation Protocols

  • Different illumination sources cannot be assigned to specific regions in this release

  • Sequential illumination of regions is not possible in this release.

  • Fixed issue where settings were grouped and moved to incorrect locations.

  • Fixed issue where empty filters displayed as magenta instead of grey.

Control of High Power Laser (Super Resolution functionality) & B-TIRF functionality

Acquisition improvements

  • Piezo z position is displayed in the multi-field table.

  • SRRF-Stream+ algorithm improvement: removal of 'donut' shaped artefact on spot-like features.

Visualization improvements

  • Easier moving and resizing of small regions. 

  • Image frame timestamp resolution increased to microseconds.

  • Taskbar icon reflects protocol status and state now including Deconvolution and Stitching.

  • Contrast level is stored per image.

Other improvements

  • Saving Z calibration information for all acquisition types using the information from microscope and Piezo position.

  • Disk Rotation On/Off  option to allow alignment of dual cameras. Dual camera alignment Instructions now included in Dragonlfy hardware manual.

  • Prevent Windows Update and User logout from shutting down Fusion while an acquisition is in progress.

  • HLE burst shutter timing threshold can be set by customer support.

Bug Fixes:

Acquisition bug fixes

  • Fix Fusion crash when running Finite Burst with Frame Average.

  • Fix for crash when editing the imaging mode.

  • Fixed crash when loading an image file from the root folder.

  • Fix to prevent crash of Fusion during long protocols with multiple channels

  • Fix ASI stage communication timeouts caused by transient errors.

  • Fix acquisition failing to resume after being paused.

  • Fix Fusion freeze when attempting a second run of a 3 channels montage.

  • Fix added to prevent images not being deleted when stopping a protocol before completion

  • Error message is now displayed if the image file path length exceeds the Windows maximum limit. Protocol won´t run until file path name is corrected (shortened).

  • Fix for Z scan size not updating correctly after maximum and minimum values are set for Analogue Piezo Z.

  • Fix for multi position acquisition slow down or hang.

  • Fix erroneous report of Zyla camera acquisition mode (finite burst protocol). Previously Fusion reported the acquisition was in external mode, while acquisition was running in internal mode.

  • Fix incorrect calculation of Finite Burst duration.

  • Fix for readout time not updating when acquisition settings changed.

  • Fix for slow acquisition start time when drift stabilisation is enabled on an IX83 microscope.

Deconvolution & stitching bug fixes

  • Fix for Fusion crashes when opening deconvolved image in split-channel MIP

  • Fix for deconvolution crashing when trying to load images where the first file in the dataset is missing

  • Fusion now prevents Windows OS shutdown while deconvolution and/or stitching processing are active

  • Fix for deconvolution slow down.

  • Fix deconvolution estimated time updates. When deconvolution is activated through the estimated time is now updated.

  • Fix for deconvolution preview window closing when clicked.

  • Fix stitching error when commas used as decimal separators.

Other bug fixes

  • Device Features for different devices of the same type are now grouped under the particular device they correspond to.

  • Fix to ensure all compatible channels can be merged or linked in dual camera simultaneous acquisition.

  • Fix for null control Unicode character in channel config files.

  • Fix for features being updated by other devices of the same type.

  • Changed the default device alias of laser controllers from "Light Source" to "Laser"

  • Improved the text error message displayed when all the fields in a table are deleted.

  • Fix for reading incorrect Objective Numerical Aperture value.

  • Fix to prevent invalid configuration files causing unsuccessful initialisation of Fusion

  • Fix for Calibration Warning not disappearing when Optical Magnification is equal for all Channels

  • Improve synchronisation of dual camera externally triggered finite burst protocols by removing delays introduced at the start of the protocol.

  • During a fast finite burst (faster than 10 Hz), leave the mechanical shutter open on laser.

  • Laser power management improved: when switching channels, lasers not in use are turned off, and then new lasers are turned on.

  • The number of log files retained has been increased to 20.

  • Added Simulated Global Shutter Clear feature for Sona cameras, to simplify synchronising the camera fire output with the confocal disk rotation.
